Methods for Analyzing Data on Cortical Activation by Voltage-Sensitive Dye

We describe methods we have found useful to characterize dynamic activity patterns over wide regions of mouse cortex as assessed by voltage-sensitive dye (VSD) imaging, primarily reflecting activity in layers 2 & 3. 

We show how multivariate statistical techniques in conjunction with Fourier analysis can characterize the changes in the spontaneous activity after sensory stimulation. We also find significant changes in how spontaneous activity patterns track the stimulus pattern. We also use novel statistical techniques to characterize the speed of activation 'fronts' and preferred directions of propagation of activation at each location in mouse cortex.
